Tengo entendido que la expresión/idiom: I´m not through with this book, significa "No he terminado de leer este libro, pero casi, casi"
¿Otras expresiones podrian ser?: I´m not through with this lesson, activity, my work,...
Por otra parte: I feel more at home doing this kind of activity. Una posible traducción puede ser: "Me siento cómodo haciendo esta actividad"

Yes, it means I've not finished doing something. However, there is no "casi, casi" element to it! It just means I've not finished yet. NB: this is American English.
The translation of the 2nd sentence is correct.
